Letrozole Tablets Description^1,2

Letrozole, an aromatase inhibitor (nonsteroidal), is used in postmenopausal women for breast cancer and for the treatment of advanced breast cancer.

Mechanism of Action

Mechanism of Action^1,2

Letrozole binds to heme group of the cytochrome P450 subunit of the aromatase (enzyme) resulting in a reduction of estrogen (estrone, estradiol and estrone sulfate) biosynthesis in all tissues. The drug decreases estrogen levels by preventing the conversion of androgens to estrogen.

Adverse Reactions

Hot flashes and sweating
Gastrointestinal (ie Nausea, vomiting, diarrhea, constipation, abdominal pain)
Musculoskeletal Disorders (ie Arthralgia, bone pain, and myalgia)
Headache
Dizziness
Bone pain

Contraindications & Precautions

Contraindications^1,2

Known hypersensitivity to letrozole or any component of the formulation
Letrozole is contraindicated during pregnancy

Precautions^1,2

Reduction in bone mineral density
Use letrozole with caution in patients with pre-existing hypercholesterolemia

Storage

Store at controlled room temperature (20°C to 25°C / 68°F to 77°F)
Protect from moisture and light
Keep in a tightly closed container
