Micronized estriol is a bioidentical form of estriol (E3)—one of the three naturally occurring estrogens in women (alongside estradiol and estrone). Estriol is the weakest estrogen, with a much lower receptor binding affinity and duration of action than estradiol (E2). Though the weakest estrogen, estriol shows potential for skin-rejuvenating properties such as restoring dermal thickness and elasticity. Estriol is most commonly used along with estradiol in postmenopausal women with estrogen-deficiency. Estriol is most commonly prepared in a transdermal cream, allowing for incremental dosing adjustments.

Aloe vera comes from the gel contained within plant leaves generally grown in hot dry geographical locations (Africa, Asia, Canary Islands). It has been used because of its pharmacological and cosmetic properties. Therapeutically, it has systemic activity as a digestive disease protectant, for its anticancer and antidiabetic properties and topically as an antioxidant and anti-inflammatory. The antioxidant and anti-inflammatory properties give the compound skin protectant capabilities and its ability to facilitate wound healing. The plant gel has 75 different compounds in it including vitamins (vitamin A, C, E, and B12), enzymes, (amylase, catalase, and peroxidase), minerals (zinc, copper, selenium can calcium) sugars (mannose-6-phosphate and glucommans), anthraquinones, fatty acids, and hormones.

Estriol acts by binding to estrogen receptors alpha (ERα) and beta (ERβ) in estrogen-responsive tissues, particularly the vaginal epithelium, bladder, urethra, and pelvic floor musculature.

Aloe vera has several major compounds including aoesin, aloin, and emodin. These compounds are thought to have protective properties when applied topically through antioxidant and anti-inflammatory activities. These properties on the skin are facilitated by aloe vera’s ability to upregulate TFGβ1, bFGF, and Vegf-A expression in fibroblasts and increase keratinocyte proliferation and differentiation by lysosomal membrane stability. It also increases type IV collagen. Aloin is believed to protect the skin through its antioxidant properties by decreasing IL-8 production, DNA damage, and reactive oxygen species formation. It exerts anti-inflammatory activity by inflammatory cell infiltration, increasing CD4+/CD8+ ratio lymphocytes, and improving epidermal thickness and collagen deposition.
